DESCRIPTION:Abstract:  B-cell lymphomas exhibit substantial genetic and cli
 nical heterogeneity\, with standard therapeutics being insufficient for ma
 ny patients. This work explores a machine learning approach to recognize d
 iagnostic entities and subgroups across three common lymphomas. Using gene
  expression from 1053 samples and a variety of machine learning methods\, 
 I selected 180 features (genes) whose expression allowed enhanced subtype 
 discrimination beyond the previously described genes. These data were then
  integrated with simple somatic mutations (SSM) and copy number variations
  (CNV)\, aiming to recapitulate the known subgroups. Among the tested mode
 ls\, a neural network integrating gene expression profiles and SSM achieve
 d the highest test accuracy (88.9%). Inclusion of CNV yielded marginal gai
 ns\, while models using only SSM+CNV alone underperformed (72.5%). This wo
 rk underscores the utility of multi-omics data and machine learning for re

DESCRIPTION:Abstract:  Netrin-1 is a secreted protein that guides axon path
 finding and modulates synapse formation and long-term potentiation in neur
 ons. Moreover\, there is a genetic association between netrin-1 and KIF1A\
 , a kinesin-3 motor protein required for axonal transport. Netrin-1 is exp
 ressed in human neurons\; however\, its intracellular distribution and tra
 fficking has not been assessed. I characterized the endogenous expression 
 of netrin-1 via immunocytochemistry and demonstrate it is localized to bot
 h the axon and dendrites of induced pluripotent stem cell (iPSC)-derived h
 uman neurons. Next\, live-cell imaging of Netrin-1-mRFP revealed robust bi
 directional axonal transport\, which is comparable to that of other cargos
  undergoing microtubule-based trafficking in vertebrate neurons. To identi
 fy the netrin-1 vesicle population\, co-localization analyses demonstrated
  that netrin-1 puncta overlap with the dense-core vesicle marker Chromogra
 nin A. Netrin-1-mRFP and Chromogranin A-GFP also undergo co-transport with
 in axon. Finally\, I demonstrated that KIF1A-associated neurological disea
 se (KAND) patient variants significantly reduce netrin-1 trafficking.\n\n\

DESCRIPTION:Abstract:  Homeodomain-interacting kinases (HIPKs) and salt-ind
 ucible kinases (SIKs) regulate cell proliferation\, apoptosis and tissue d
 evelopment. In Drosophila melanogaster\, the Hipk ortholog (dHipk) has bee
 n shown to promote overgrowth and tumorigenic phenotypes\, and when co-exp
 ressed with Sik2 or Sik3\, the tumour-like overgrowth significantly increa
 ses. This project investigates whether this synergistic interaction transl
 ates to human homologs in both Drosophila and human cell lines. Using tran
 sgenic flies and cell lines (MCF7\, HEK293T)\, the phenotypes related to t
 issue overgrowth\, developmental delay\, apoptosis and migration were anal
 yzed. In flies\, the coexpression of HIPK1/2 with constitutively active Si
 k3 resulted in developmental delay\, reduced viability and severe wing def
 ects\, supporting an oncogenic synergy. In human cells\, this coexpression
  led to varied effects\, including reduced proliferation and enhanced apop
 tosis\, with HIPK2 having a stronger pro-apoptotic synergy. These results 
 support a context-dependent HIPK-SIK3 cooperation in the context of tumori

DESCRIPTION:Abstract: To survive\, cells and organisms must be capable of a
 dapting to changing environmental conditions. My lab studies an unconventi
 onal stress response pathway that utilizes physiological amyloid aggregati
 on to maintain viability. Here\, exposure to stimuli (e.g.\, heat shock an
 d acidosis) triggers the formation of solid-like biomolecular condensates\
 , termed amyloid bodies or A-bodies. These structures recruit and inactiva
 te a large family of cellular proteins\, which adopt an amyloid-like confo
 rmation upon entering the subnuclear foci. Interestingly\, divergent stimu
 li sequester unique A-body constituents\, providing cells with a mechanism
  to tailor the metabolic response to specific environmental changes. While
  A-bodies share many biophysical characteristics with the pathological amy
 loids associated with common neurodegenerative disorders (e.g.\, Alzheimer
 ’s and Parkinson’s disease)\, a unique feature A-body biology is that 
 these physiological aggregates can rapidly disassemble upon returning cell
 s to normal growth conditions. Mapping the cellular signaling cascades reg
 ulating with this process has demonstrated that stress-specific post-trans
 lational modifications of the heat shock chaperone machinery is critical f
 or amyloid disaggregation. Thus\, we hope that by understanding the cellul
 ar mechanism regulating assembly and disassembly of these functional amylo
 ids we may be able to uncover interesting new therapeutic avenues for comb

DESCRIPTION:Abstract:  Cyclin dependent kinases (CDKs) regulate cell cyclin
 g\, gene expression\, and illustrated\nrecently\, mitochondrial dynamics. 
 Depleting Drosophila cdk8\, the ortholog to human CDK8 and CDK19\, induces
  various phenotypes that can be rescued by expressing CDK19WT. Heterozygou
 s dominant missense mutations in CDK19 have been characterized in patients
  with developmental and neuromuscular deficiencies\, termed Developmental 
 Epilepsy and Encephalopathy-87 (DEE87) syndrome. With conflicting views on
  the functional nature of variants\, Y32H and T196A\, our hypotheses aimed
  to assess the effects of ubiquitous variant expression on fly health\, mu
 scle\, and motor function using the GAL4/UAS system. Y32H rescues several 
 cdk8 knockdown phenotypes possibly by gain of function compensation\, whil
 e T196A failed to rescue\, possibly by loss of kinase activity/dominant ne
 gative effect. Dietary NACA supplementation improves T196A phenotypes. Our

DESCRIPTION:Abstract:\nResearch in the Braid lab focuses on human mesenchym
 al stromal cells (MSCs)\, a fascinating and still enigmatic population of 
 progenitor cells that function as signaling hubs to coordinate immune and 
 progenitor cell behaviour. MSCs are rapidly advancing as clinical treatmen
 ts for numerous diseases\, owing to their ability to reduce inflammation a
 nd promote regeneration. However\, the mechanisms used by MSCs exert these
  effects remains unclear. Our group is particularly interested in understa
 nding how MSCs contribute to their cellular communities to influence devel
 opment\, tissue homeostasis and repair. We apply this knowledge to develop
  new resources for modeling human tissues in vitro and to enhance the effe
 ctiveness of MSC-based therapies. In this seminar\, I will discuss how our
  team is characterizing and leveraging the niche-organizing properties of 
 MSCs through three research pillars: (1) Developing new biomaterials\, fun
 ctional assays and high-fidelity tissue models using human matrix componen
 ts\, (2) Investigating how lysosomal dysfunction in MSCs drives aging and 
 disease\, and (3) Characterizing and leveraging new molecular players in M

DESCRIPTION:Abstract:\nEach year\, approximately three million newborns die
  from sepsis worldwide\, with ~75% of all deaths under-five years occurrin
 g within the first week of life. Sepsis is a global concern\, especially t
 o young infants who are at highest risk of mortality. However\, there lack
 s a quick and reliable way to identify the infectious microbes\, resulting
  in babies being over- or under-treated with antibiotics that may contribu
 te to resistance and depletion of limited resources. To tackle this challe
 nge\, our group is applying systems biology approaches and advanced omics 
 analyses within a Global Health context to: (1) define host molecular mark
 ers in bacterial sepsis using genome-wide transcriptomic analyses and mach
 ine learning\; (2) identify potential virulence factors in the causal bact
 erial pathogens using microbial genome-wide association studies\; and (3) 
 apply cutting-edge long-read Nanopore 16S sequencing to develop vaccines a
 nd improve pathogen identification. Ultimately\, our goal is to understand
  why newborns are highly susceptible to infections during their first week
  of life\, and apply advanced omics strategies to develop real-world appli

DESCRIPTION:Abstract: Macrophages are tissue-resident sentinels of the immu
 ne system. They clear unwanted material by phagocytosis\, regulate inflamm
 ation and activate lymphocytes through antigen presentation. They are ofte
 n the most abundant immune cells\, both in healthy peripheral tissues and 
 in tumor microenvironments. Thus\, various immunotherapies have been devel
 oped to harness the functions of macrophages against cancer. These include
  antigen-targeting monoclonal antibodies\, which have led to significant i
 mprovements in the treatment of some malignancies. Macrophages recognize a
 ntibody-bound cancer cells through Fc-γ receptors\, which activate signal
 ing pathways that culminate in the engulfment of cancer cells by phagocyto
 sis. However\, engulfing entire living cells is remarkably challenging and
  is limited by immune checkpoints and physical constraints that prevent it
 s occurrence in normal conditions. To study the constraints that limit the
  efficacy of cellular phagocytosis\, we developed genetic engineering and 
 quantitative live cell microscopy approaches. We found that phagocytosis o
 f cancer cells involves co-receptors that tightly fasten macrophages to th
 eir target. Moreover\, our work revealed that malignant cells can take adv
 antage of this mechanism and evade phagocytosis through frequently occurri
 ng mutations that affect their biophysical properties. These findings open
  new perspectives on strategies to improve the phagocytic capability of ma

DESCRIPTION:Abstract:\nThe ribosome translates information provided by the 
 genetic code into functional proteins by forming peptide bonds between ami
 no acids. The ribosomal RNA (rRNA) that catalyses this reaction is largely
  conserved across all domains of life and resistant to change. To engineer
  the ribosome\, the Willi lab uses in vitro and in vivo methods to build n
 ew ribosomes from scratch without sacrificing cell viability. With these s
 ynthetic biology methods we seek to explore both the past and future of th
 e ribosome.\nMultiple models of the origin and evolution of the translatio
 n system have resulted from molecular-level modelling and phylogenetic ana
 lysis of rRNA. These models posit that the present-day rRNAs evolved by ac
 cretion of expansion segments from the two functional cores\, and in later
  phases co-evolved with ribosomal proteins and translation factors. We rem
 oved several of the proposed ancestral expansion segments from the modern 
 rRNA and assembled the resulting proto-ribosomes for testing of translatio
 nal fitness. Short linkers to heal the truncated sites were optimized usin
 g ribosome display. This resulted in a range of translationally active pro
 to-ribosomes missing stretches of 20-166 nt each\, as well as combinations
  of deletions\, resulting in up to 40% smaller rRNA and loss of ribosomal 
 proteins.\nUsing the same techniques\, we are also engineering the ribosom
 e to expand beyond the "normal" protein translation it evolved to do and u
 nlock entirely new modes of polymer synthesis. Using in vivo selection\, w
 e altered the decoding mechanism to expand coding space beyond the standar
 d triplet base codons and efficiently accept quadruplet-tRNA. Furthermore\
 , in vitro evolution was used to create custom specialized ribosomes for e
 nhanced production of difficult proteins and introduction of non-canonical

DESCRIPTION:Abstract: In our brain\, every decision begins with a neuron co
 mmunicating a signal through a synapse. During my postdoctoral studies\, I
  discovered a novel transcriptional program\, controlled by neuronal activ
 ity\, which drives synaptogenesis in C. elegans dopaminergic neurons. This
  work was among the first to demonstrate a link between activity and devel
 opmental transcription factors to sculpt synapse formation in vivo.  In my
  new lab at UBC\, we will leverage the molecular and cell biology tools I

DESCRIPTION:Abstract: RNA molecules perform a staggering array of functions
  in all kingdoms of life\, executing both the core steps of protein synthe
 sis and an ever-growing repertoire of noncoding roles essential to cellula
 r physiology. In humans\, RNAs are also prolific organizers of subcellular
  compartmentalization\, scaffolding membraneless organelles and chromatin 
 domains that collectively orchestrate cells' gene-expression\, metabolic\,
  and stress-response programs. Yet efforts to decipher the molecular mecha
 nisms underlying these important functions have long been challenged by a 
 lack of robust technologies for discovering the other factors (proteins\, 
 RNAs\, genomic loci) with which an RNA interacts within its native cellula
 r context. Technologies that overcome this hurdle would thus provide power
 ful insights into fundamental RNA-regulatory pathways in across organisms\
 , and support the development of new classes of RNA-targeting therapeutics
 .\n                My research program seeks to meet this need by developi
 ng advanced\, yet accessible Chemical Biology tools for dissecting RNA fun
 ction in situ\, and using these tools to understand how RNA shapes nuclear
  architecture during human development and disease. Toward this goal\, we 
 have recently developed O-MAP\, a versatile and straightforward method for
  mapping the proteins\, RNAs\, and genomic loci within the subcellular mic
 roenvironment surrounding nearly any RNA of interest—with unmatched dept
 h and spatial precision. O-MAP uses RNA-FISH-like oligonucleotide probes t
 o deliver proximity-biotinylating enzymes to a target RNA in situ\, enabli
 ng nearby molecules to be enriched by streptavidin pulldown. This induces 
 exceptionally precise biotinylation that can be easily optimized and porte

 ting nascent pre-mRNAs\, we demonstrate that O-MAP can "dissect" the uniqu
 e molecular architecture of individual nuclear compartments at single-gene

DESCRIPTION:Abstract: Plasmids play a critical role in disseminating antibi
 otic resistance genes yet their host association within microbial communit
 ies remains challenging due to difficulties in binning plasmids\, which of
 ten have distinct sequence characteristics compared to chromosomal DNA. Hi
 -C sequencing shows promise by crosslinking DNA in physical proximity\, su
 ch as plasmid and chromosomal DNA from the same cell. In this study I deve
 loped HiCPlas\, a Nextflow pipeline for binning and reconstructing plasmid
 s from Hi-C metagenomic samples. Using a simulated community\, HiCPlas ass
 ociated up to 40 out of 65 plasmids with their hosts\, far surpassing the 
 two recovered by traditional binning tools. Applying this pipeline to two 
 public metagenomics datasets\, it identified over 200 plasmid-host associa
 tions including 40 plasmids carrying antimicrobial resistance genes\, 34 p
 lasmids outside reported host ranges and several putative transmission eve
 nts. These results demonstrate the potential of Hi-C metagenomics and the 
 HiCPlas pipeline for resolving plasmid dynamics at the community level.\n\

DESCRIPTION:Abstract: A long-standing question in cell biology is how lipid
 s are distributed from their main site of synthesis – the endoplasmic re
 ticulum (ER) – to other organelles to support their biogenesis and speci
 alized functions. Recent studies suggest that lipid transport proteins (LT
 Ps)\, which act at membrane contact sites where organelles come into close
  proximity\, play a central role in mediating inter-organelle lipid exchan
 ge. Despite their importance\, in vivo evidence directly linking the loss 
 of specific transport pathways to defined lipid trafficking defects remain
 s limited. This knowledge gap largely stems from the lack of robust assays
  to monitor lipid transport dynamics within living cells.\nIn this talk\, 
 I will describe synthetic biology-based strategies we have developed to ad
 dress this challenge\, including a new tool called METALIC (Mass Tagging
 –Enabled TrAcking of Lipids In Cells)\, which allows direct tracking of 
 inter-organelle lipid flux in vivo. Using METALIC-based approaches\, we ha
 ve uncovered previously unknown routes of lipid transport and demonstrated

DESCRIPTION:Abstract:\nA mechanistic basis for non-small cell lung cancer (
 NSCLC) initiation in never smokers\, a disease with high frequency EGFR mu
 tations (EGFRm)\, is unclear. Air pollution particulate matter (PM) is kno
 wn to be associated with the risk of NSCLC\, however a direct cause and me
 chanism remain elusive. We analysed 463\,679 individuals to address the as
 sociations of increasing 2.5um PM (PM2.5) concentrations with cancer risk.
  We performed ultra-deep profiling of 247 normal lung tissue samples\, ana
 lysed normal lung tissue from humans and mice following exposures to PM\, 
 and investigated the consequences of PM in mouse lung cancer models. Incre
 asing PM2.5 levels are associated with increased risk of EGFRm NSCLC in En
 gland\, S.Korea and Taiwan. 18-33% of normal lung tissue samples harbour d
 river mutations in EGFR and KRAS in the absence of malignancy. PM promotes
  a macrophage response and a progenitor-like state in lung epithelium harb
 ouring mutant EGFR. Consistent with PM promoting NSCLC in at-risk epitheli
 um harbouring driver mutations\, PM accelerates tumourigenesis in three EG
 FR or KRAS driven lung cancer models in a dose-dependent manner. Finally\,
  we uncover an actionable inflammatory axis driven by IL1B in response to 
 PM\, in agreement with reductions in lung cancer incidence with anti-IL1ß
  therapy. These results shed light on the etiology of EGFRm lung cancer an
 d suggest that oncogenic mutations may be necessary but insufficient for t
 umour formation. These data reveal a mechanistic basis for PM driven lung 
 cancer providing an urgent mandate to limit air pollution. Importantly\, t
 he observation of a reduction in pollution-induced tumour growth upon anti
 -IL1ß treatment suggested that tumorigenesis may be prevented.\n\nSpeaker

DESCRIPTION:Abstract:\nOxygen is essential for cellular survival and functi
 on\, yet many diseases—including lung disease\, stroke\, and cancer—de
 prive tissues of this critical resource. This oxygen deprivation\, or hypo
 xia\, drives much of the tissue damage characteristic of these conditions.
  Understanding how cells and tissues adapt their metabolism to survive low
  oxygen is therefore a fundamental question in biomedical research.\n\nWe 
 are addressing this question using Drosophila as a study system. Because t
 hey naturally grow and develop in decaying food\, an environment character
 ized by low local oxygen levels\, flies have evolved remarkable tolerance 
 to low oxygen. Our work focuses specifically on how inter-organ communicat
 ion orchestrates whole-body adaptation to low oxygen. In my talk\, I will 
 describe our recent findings showing how specific tissues sense hypoxia an

 n in E. coli bacteria in response to diverse stimuli in the environment an
 d the host and are essential for stress response and virulence. Most sRNAs
  regulate mRNA expression by directly base pairing with complementary site
 s in the target mRNA with the help of the chaperone protein Hfq. sRNAs and
  Hfq must rapidly search hundreds of candidate mRNAs for matched targets w

DESCRIPTION:Abstract: This presentation has two parts.\n\nStructural variat
 ions (SVs) greatly contribute to human genome diversity. Additionally\, so
 matic SVs in tumor genomes have been found to play a critical role in the 
 development of cancer. We present a deep learning model designed and train
 ed for SV identification\, to enable the detection of a full spectrum of s
 ubclonal SV frequencies and the detection of somatic SVs from matched tumo
 r-normal genome pairs.\n\nThe assessment of fertilized human embryos is a 
 crucial step in IVF\, traditionally performed by embryologists but now bei
 ng enhanced by deep learning-based AI techniques. Current predictive model
 s for assessing embryo quality and chromosomal abnormalities (ploidy) have
  not effectively utilized time-lapse imaging\, limiting accuracy and preve
 nting the identification of key developmental time points that could impro
 ve predictions. To address this\, our lab developed and AI foundation mode
 l and various predictive models and compared their performance across diff
 erent developmental stages\, with the results to be presented in the secon

DESCRIPTION:Abstract: The intestinal microbiome plays a critical role in im
 mune development and regulation\, yet most research has focused on bacteri
 al communities. Emerging evidence suggests that non-bacterial members of t
 he microbiome also have profound effects on host immunity. My work focuses
  on a commensal protozoan as a previously underappreciated regulator of im
 mune responses. Colonization with this protozoan activates a gut-initiated
  immune pathway that reshapes immune responses at distant sites\, includin
 g the lung\, where it exacerbates allergic airway inflammation while enhan
 cing protection against infection. These effects are mediated in part by g
 roup 2 innate lymphoid cells (ILC2s)\, a specialized population of tissue-
 adapted immune cells that integrate microbial and tissue-derived signals t
 o help establish the immune tone of different organs. Together\, my work h
 ighlights how non-bacterial microbes and tissue-specialized immune cells c

DESCRIPTION:Abstract: Viruses have evolved diverse strategies to take over 
 the host translation machinery such as the ribosome to direct viral protei
 n synthesis. Revealing these strategies have uncovered non-canonical modes
  of translation\, illuminating the diverse ways protein synthesis can occu
 r. My lab has examined two such mechanisms. One\, we have examined an unpr
 ecedented viral RNA structure that can recruit ribosomes directly – no p
 roteins required\, no initiator Met-tRNAi required – and can initiates t
 ranslation from a non-AUG codon. I will discuss how we have used bioinform
 atic mining of viral genome databases to identify novel “all-in-one” f
 actorless translation RNA elements and hidden alternative reading frames d
 riven by these elements\, thus revealing the evolutionary framework of thi
 s RNA-driven mechanism. Two\, we have recently studied how an ~20 amino ac
 id viral peptide called 2A can mediate stop-go translation that leads to p
 roduction of separate proteins from a single open reading frame. Despite i
 ts widespread use in research and therapeutic settings\, the mechanism und
 erlying this 2A peptide mechanism remains poorly understood. I will discus
 s how we have developed a sensitive 2A-sensing reporter for high-throughpu
 t screening of novel functional 2A peptides and comprehensive mutagenesis\
 , thus revealing the molecular functional rules and potential mechanisms o

DESCRIPTION:Abstract: The next generation of antibody therapeutics hold the
  potential to transform global health outcomes. Despite this promise\, ant
 ibody discovery remains biologically complex\, resource-intensive\, and vu
 lnerable to failure at multiple stages. At AbCellera\, diverse and cutting
 -edge technologies—including high-throughput single B-cell screening—h
 ave been integrated to accelerate discovery timelines\, increase molecular
  diversity\, and generate candidates with strong developability profiles\,
  thereby de-risking downstream development.\n\nIn this talk\, we will prov
 ide an overview of the full discovery workflow\, from immunization and B-c
 ell screening to antibody production\, validation\, and early developabili
 ty assessment. We will highlight the central role of protein science acros
 s these phases\, with emphasis on high-throughput antibody expression and 
 characterization\, and structural biology. These biochemistry-intensive ca
 pabilities form an engine of innovation that aims to increase the probabil

DESCRIPTION:Abstract: Human induced pluripotent stem cells (hiPSCs) have pr
 oven invaluable for human disease modeling and translation. Additionally\,
  Self-assembling human organoid models promise to enable studies of human 
 organogenesis in unprecedented detail. Up to date\, several groups generat
 ed self-assembling human organoids such as neural\, liver\, intestinal\, t
 hymic and cardiac organoids. These novel human in vitro systems are critic
 al to overcome current limitations of widely used animal models\, which do
  not reproduce human organ development and fail in validation of molecular
  drug targets. In fact\, only 8% of clinical cancer trials successfully tr
 anslate from animals to humans. However\, challenges with quality\, cellul
 ar specification and maturity\, inter-batch consistency\, cryopreservation
  and scale remain\, reducing experimental reproducibility and clinical tra
 nslation of currently used cell and organoid models. Additionally\, these 
 organoids do not fully recapitulate multilineage development and cellular 
 crosstalk present in human organs. The goal of my research group is to adv
 ance the state-of-the-art in modeling human organ development and disease.
  Thus\, we recently reported using 3D suspension systems to enhance qualit
 y\, reproducibility\, and yield of hiPSC-derived cardiomyocytes (hiPSC-CMs
 ). In the same study\, I showed that this approach also generated the firs
 t reported fully suspension-derived cardiac organoid. My current unpublish
 ed data shows that with further refinement I am able to use suspension cul
 ture to induce formation of multilineage cardiac\, neural\, gastric and in
 testinal organoids. Further\, using the same methodology and chemical cues
  I can generate mixed-lineage organoids (e.g. cardiac-neural\, intestinal-
 neuronal) and multi-organ organoids\, resembling “organs on a string”.
  These studies will provide the necessary framework and first steps for ge
 nerating new human in vitro models reproducing the complex cellular intera
 ctions that are necessary to guide human organ development\, function and 
 disease\, while providing a more relevant testbed for future human medicin

DESCRIPTION:Abstract: Arthropod-borne\, positive-sense RNA viruses are majo
 r human pathogens that threaten global health. Effective antiviral strateg
 ies require a detailed understanding of the intracellular dynamics and hos
 t interactions of these viral genomes. We have developed dual-labeled Zika
  and chikungunya viruses engineered with the Mango-II RNA aptamer\, enabli
 ng both RNA pulldown and real-time\, high-resolution imaging in live cells
 . By pairing RNA imaging with visualization of viral replication proteins\
 , we revealed the spatiotemporal dynamics of replication complexes and RNA
  molecules. Comprehensive proteomic analyses identified over 1\,000 high-c
 onfidence host factors interacting with viral RNA\, with 355 corroborated 
 from existing datasets. Notably\, we discovered that the spliceosome facto
 r SFPQ is a shared interactor\, and its depletion impairs RNA production f
 or both viruses. Confocal microscopy confirmed intracellular colocalizatio
 n of SFPQ with ZIKV and CHIKV RNA. We extended our approach to human place
 ntal cells\, identifying more than 1\,200 RNA-protein interactions\, and u
 tilized super-resolution microscopy to visualize viral RNA and host protei
 ns. Additional pulldown assays using a streptavidin-tagged ZIKV capsid pro
 tein identified eight overlapping host proteins\, including cytoskeletal p
 roteins PLEC\, IQGAP1\, and TLN1. Finally\, we demonstrate the versatility
  of Mango-II tagging by generating a tagged chikungunya virus construct fr
 om the Togaviridae family. Together\, these results establish Mango-II apt
 amer tagging as a powerful platform for dissecting the spatiotemporal dyna
 mics and host interactions of positive-strand RNA viral genomes\, offering

DESCRIPTION:Abstract: Muscle stem cells\, also known as satellite cells\, a
 re essential for muscle regeneration and for maintaining muscle homeostasi
 s and regenerative capacity throughout life. Increasing evidence supports 
 a contributing role for satellite cells in Duchenne muscular dystrophy (DM
 D)\, a lethal degenerative muscle disease. DMD is caused by mutations in D
 MD that result in the loss of dystrophin protein. Using single‑cell RNA 
 sequencing\, we found that satellite cells in DMD exhibit altered stem cel
 l heterogeneity and cell‑fate trajectories. DMD satellite cells were ske
 wed toward myogenic progenitor fates\, and a significant proportion were f
 ound within a DMD‑enriched cluster. We also identified dysregulated mole
 cular pathways in DMD satellite cells\, including enhanced apoptosis\, inc
 reased senescence\, and dysregulated cellular autophagy. Importantly\, our
  single‑cell transcriptomic analyses and in vivo regeneration assays dem
 onstrate that DMD satellite cells express an impaired myogenic differentia
 tion gene signature and are stalled in their differentiation capacity. Ove
 rall\, our findings provide insight into the molecular pathways that are d
 ysregulated in DMD satellite cells and suggest molecular targets that may

DESCRIPTION:Abstract: Bacteria compete intensely with neighbouring microbes
  for access to nutrients and space\, often deploying toxic proteins to eli
 minate competitors. In this seminar\, I will describe our discovery of a n
 ew family of antibacterial proteins (ABPs) that function as interbacterial
  toxins capable of killing diverse Gram-positive bacteria. ABPs are produc
 ed as protoxins that are proteolytically activated by dedicated co-exporte
 d serine proteases. Once activated\, these toxins enter target cells in a 
 process driven by the proton motive force and rapidly inhibit essential ce
 llular processes. Biochemical and structural studies reveal that ABP toxin
 s encompass multiple enzymatic activities and that purified toxins act as 
 potent protein antibiotics against diverse Gram-positive bacteria. Togethe
 r\, these findings uncover a previously unrecognized strategy by which bac
 teria deploy diffusible protein antibiotics and suggest that ABP systems r
 epresent a rich and largely unexplored source of antimicrobial molecules.\

DESCRIPTION:Abstract: Using pharmacogenomics\, the study of how genetic var
 iability contributes to individual drug responses\, we are identifying gen
 etic factors that can help predict an individual’s response to opioid-ba
 sed pain relievers. Following a pipeline from genetic discoveries to predi
 ctive genetic testing\, our work will help select the safest and most effe
 ctive pain relievers for vulnerable children\, such as preterm infants and
  children with cancer. Specifically\, we are working to 1) discover robust
  genetic factors that influence variability in opioid responses in childre
 n\, 2) validate discovered genetic factors using a C. elegans pain managem
 ent model\, and 3) implement precision medicine initiatives through the de
 velopment of pharmacogenomic testing strategies to improve outcomes. Toget
 her\, this will allow for more individualized risk-benefit decisions for p
 ain management in children\, while also contributing to the discovery of n
